Juvenile idiopathic arthritis (JIA) is a chronic, inflammatory condition affecting children's joints, causing pain, swelling, stiffness, and limited motion range. An imbalance in immune cells, influenced by genetic and environmental factors, contributes to JIA pathogenesis.

A recent study by researchers from the Children's Hospital of Chongqing Medical University analyzed immune cells' single-cell transcriptomes in JIA patients, focusing on sJIA and non-sJIA subtypes to understand the immune response in JIA progression.

The study used droplet-based single-cell RNA sequencing on peripheral blood mononuclear cells from 14 JIA patients and four healthy controls. Results showed increased levels of certain cells in JIA, with a more pronounced pro-inflammatory response in sJIA compared to non-sJIA.

Analysis of CD4+ and CD8+ cell transcriptomic profiles revealed differences in gene expression related to apoptotic signaling and inflammatory responses between sJIA and non-sJIA. B-cell response to IFN was stronger in sJIA, indicating potential factors contributing to different clinical manifestations.

CellChat analysis showed abundant interactions between various immune cells in both sJIA and non-sJIA cases. A new ligand-receptor pair, MIF (CD74-CXCR2), was identified in sJIA, suggesting its potential as an indicator to distinguish sJIA from non-sJIA.

This study, the first to characterize immune cells' single-cell transcriptomic profile in different JIA subtypes, has significant implications for precise diagnosis, classification, and personalized treatment strategies for JIA patients.
